초록(요약문)
현장진단(point-of-care testing, POCT)은 복잡한 실험실 장비 없이 현장 에서 신속하고 간단하게 정량 분석을 수행할 수 있는 기술을 필요로 한다. 대표적으 로 ELISA 및 PCR과 같은 기존 진단법은 높은 분석 성능을 제공하지만, 긴 분석 시 간, 반복적인 세척 과정, 대형 장비의 사용 등으로 인해 현장 적용에 한계가 있다. Lateral flow assay(LFA)는 빠르고 간편하게 사용할 수 있지만, 주로 정성 또는 반 정량 분석에 머무르는 경우가 많아 정량 진단에는 제한이 있다. 따라서 기존 진단법 의 복잡성과 LFA의 낮은 정량성을 보완할 수 있는 휴대형 정량 진단 기술의 개발이 요구된다. 본 연구에서는 흡입 기반 유체 구동, 미세유체 혼합, 광학 검출 및 무선 신 호 전송 기능을 통합한 휴대형 미세유체 광학 바이오센싱 플랫폼을 개발하였다. 본 플랫폼은 압타머 매개 금 나노입자(AuNPs) 응집 반응을 기반의 검출 메커니즘을 사 용하였고 별도의 세척 과정 없이 바이오마커를 검출할 수 있도록 설계되었다. 압축 스프링 기반 수동 흡입 펌프를 이용하여 외부 전기 펌프 없이 시료와 시약을 이송하 였고 serpentine 미세채널을 통해 시약 혼합을 촉진하였다. 또한 650 nm 레이저 다 이오드와 포토다이오드를 이용하여 금 나노입자 응집에 따른 산란광 변화를 실시간으 로 측정하고 이를 바이오마커 농도 분석에 활용하였다. 개발된 플랫폼의 성능은 트롬 빈과 SARS-CoV-2 S1 단백질을 모델 바이오마커로 사용하여 검증하였다. 약 5분 이내에 정량 분석을 수행하였고 트롬빈 0.5 nM 및 SARS-CoV-2 S1 단백질 0.4 nM의 검출한계를 달성하였다. 또한 수동 흡입 펌프는 변동계수 5% 이하의 재현성 있는 유체 구동 성능을 보여, 사용자 의존성이 낮은 안정적인 유체 제어 가능성을 확 인하였다. 본 연구는 수동 흡입 기반 미세유체 구동과 금 나노입자 기반 산란광 검출 시스템을 통합함으로써, 신속하고 세척 과정이 필요 없는 휴대형 정량 현장진단 플랫 폼의 가능성을 제시하였다. 또한 LFA의 제한적인 정량성을 보완하여 현장에서 신속 한 1차 선별검사와 초기 판단을 지원하는 데 활용될 수 있다. 향후에는 공압 멤브레 인 밸브 기반 자동화와 CNN 기반 이미지 분석을 적용하여 사용자 조작을 줄이고 정 량 분석 성능을 향상시킬 수 있을 것으로 기대된다.

Point-of-care testing (POCT) requires technologies that can provide rapid, simple, and quantitative analysis at the site of sample collection without complex laboratory equipment. Conventional diagnostic methods, such as enzyme-linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR), offer high analytical performance but are limited for on-site applications due to long analysis times, repeated washing steps, and the need for bulky instruments. Lateral flow assays (LFAs) are rapid and easy to use; however, they are often limited to qualitative or semi-quantitative analysis. Therefore, portable quantitative diagnostic technologies are needed to complement the complexity of conventional diagnostic methods and the limited quantification capability of LFAs. In this study, a portable microfluidic optical biosensing platform was developed by integrating suction-driven fluid actuation, microfluidic mixing, optical detection, and wireless signal transmission. The platform uses an aptamer-mediated gold nanoparticle (AuNP) aggregation-based detection mechanism and was designed to detect biomarkers without additional washing steps. A spring-driven manual suction pump was used to transport samples and reagents without an external electric pump, while a serpentine microchannel promoted reagent mixing. In addition, a 650 nm laser diode and a photodiode were used to measure real-time changes in scattered light induced by AuNP aggregation, and the measured optical signals were used for biomarker concentration analysis. The performance of the developed platform was validated using thrombin and SARS-CoV-2 S1 protein as model biomarkers. Quantitative analysis was achieved within approximately 5 min, with limits of detection of 0.5 nM for thrombin and 0.4 nM for SARS-CoV-2 S1 protein. The manual suction pump also showed reproducible fluid actuation with a coefficient of variation below 5%, confirming stable fluid control with reduced user dependence. This study demonstrates the feasibility of a rapid and wash-free portable quantitative POCT platform by integrating suction-driven microfluidic actuation with AuNP-based scattering detection. The platform can complement the limited quantitative capability of LFAs and support rapid first-line screening and preliminary decision-making in on-site diagnostic environments. In future work, pneumatic membrane valve-based automation and CNN- based image analysis are expected to further reduce user operation and improve quantitative analytical performance.
