초록(요약문)
Standard redox potentials are critical thermodynamic descriptors for understanding and predicting complex toxicological endpoints, such as skin irritation and severe corrosion. Many xenobiotics induce direct cytotoxicity and disrupt epidermal redox homeostasis by acting as intrinsic oxidants or undergoing metabolic oxidation to generate reactive species. Consequently, their oxidation potentials are directly linked to their toxicological activation. Despite their mechanistic importance, experimental redox potentials are very limited across a broad chemical space, thereby limiting the development of robust predictive models. In this study, a large-scale dataset of standard oxidation potentials for 2,770 compounds was generated using Density Functional Theory (DFT). Focusing on the neutral-to-cation transition as a proxy for electrophilic reactivity, calculations employed a three-step Multilevel Ansatz. Geometries and thermal corrections were obtained at the B3LYP-D3(BJ)/Def2-SVPP level, while single-point electronic and solvation energies were evaluated at the B3LYP-D3(BJ)/Def2-TZVPP level using the Conductor-like Polarizable Continuum Model (CPCM). Reaction free energies were evaluated via the Born-Haber thermodynamic cycle and converted to the standard hydrogen electrode (SHE) scale. A perturbation protocol was applied to correct saddle point structures. During this process, 577 molecules from an initial 3,347-compound library were excluded due to SCF convergence failures, spin contamination, or persistent imaginary frequencies. This filtering process ensured the thermodynamic validity of the final 2,770-compound dataset. An automated Python pipeline was deployed to extract data from over 6,000 calculation logs. Validation against 311 experimental benchmark compounds from the ROP313 database yielded an overall Mean Absolute Error (MAE) of 0.363 V, with the organic subset demonstrating an MAE of 0.259 V. The computed standard oxidation potentials serve as thermodynamic descriptors for electrophilic reactivity, without reliance on an arbitrary electron acceptor. This dataset provides a feature set for developing machine learning (ML) models and mechanistically grounded risk assessments in non-animal chemical safety.

표준 산화환원 전위는 피부 자극성 및 부식성과 같은 복잡한 독성학적 종말점을 이해하고 예측하기 위한 핵심적인 지표이다. 피부 독성을 유발하는 수많은 이물질은 체내에서 산화되어 반응성 중간체를 생성하고 세포막을 파괴하므로, 분자의 산화 전위는 독성 활성화 메커니즘과 직결된다. 그러나 광범위한 화학 공간에 대한 실험적 전위 데이터의 부족은 신뢰도 높은 예측 모델 개발의 근본적 한계로 작용해왔다. 본 연구에서는 밀도범함수이론을 활용하여 2,770종의 화합물에 대한 대규모 표준 산화 전위 데이터베이스를 구축하였다. 생체 내 친전자성 반응을 반영하는 ‘중성에서 양이온으로의 산화 과정’에 집중하여 3단계의 다중 레벨 접근법을 적용하였다. 구조 최적화 및 열적 보정은 더블제타 수준에서, 단일점 전자 에너지 및 수용액상 용매화 에너지는 편광 가능 연속체 모델 기반의 트리플제타 수준에서 정밀하게 평가되었다. 또한, 섭동 알고리즘을 도입하여 안장점 구조를 국소 최솟값으로 안정화하였다. 이러한 품질관리 과정을 통해, 초기 3,347종 중 수렴 실패나 스핀 오염, 지속적인 안장점 구조를 보인 577종을 제외하여 최종 2,770종 데이터의 열역학적 타당성을 확인하였다. 6,000여 개의 로그 파일 데이터는 자동화된 파이썬 파이프라인을 통해 일괄적으로 추출되었다. 311종의 벤치마크 데이터를 통한 검증 결과, 전체 평균 절대 오차는 0.363 볼트였으며, 특히 유기 화합물 하위 그룹에서는 0.259 볼트의 정확도를 나타내어 계산된 전위가 실험값과 잘 일치함을 확인하였다. 본 연구에서 도출된 산화 전위 데이터는 물질 고유의 친전자성을 평가하는 유효한 열역학적 지표이다. 구축된 데이터베이스는 피부 독성 메커니즘에 기반한 위해성 평가 및 동물 대체 시험법을 위한 기계학습 모델의 주요 입력 특징으로 활용될 수 있다.
