Enhanced Sonodynamic Therapy using Focused Ultrasound-Activated Oxygen Carrying Nanodroplet for the Treatment of Hypoxic Tumors

Recently, sonodynamic therapy using sonosensitizers encapsulating nanoparticles has been intensively investigated, due to the limitation of photodynamic therapy of the light penetration depth. However, the hypoxic condition of cancer tissue limits the effectiveness of sonodynamic therapy. In this study, we loaded decafluoropentane (DFP) with high oxygen carrying capability in the core of a phospholipid based nanodroplet. Also, for sonodynamic therapy, verteporfin was loaded in the nanodroplet. When the nanodroplets are delivered to the cancer tissue, focused ultrasound (FUS) is irradiated for the evaporation of the DFP and the cavitation of nanodroplets, while temporarily generating heat and releasing excess oxygen to overcome hypoxic condition. The FUS-activated verteporfin loaded nanodroplet generated sufficient amounts of ROS and demonstrated improved sonodynamic therapy efficacy in in vitro and in vivo experiments. In conclusion, the DFP and verteporfin loading nanodroplet showed sufficient potential in cancer therapy by overcoming the hypoxia condition of tumor environment and the limitations of PDT.
