High Cell Density Fed-batch Cultivation of Methylobacterium Extorquens AM1 for Formate Dehydrogenase Production

Methylobacterium extorquens AM1 expresses formate dehydrogenase 1 (MeFDH1) as the key enzyme responsible for converting carbon dioxide to formate, a fundamental step to building chemical platforms from greenhouse gases in a greener way. In this study, a fed-batch cultivation strategy was developed for the mass production of MeFDH1 from the homologous recombinant Methylobacterium extorquens AM1. Methanol was used as a feeding substrate with the designed composition. When the carbon source was depleted, pH and DO increased sharply, and fed-batch operations were developed based on this phenomenon. Through medium optimization and the supply of carbon and nitrogen source using the pH and DO data, the high cell density culture of AM1 cells could be obtained, thus allowing higher FDH enzyme production. At the derived condition, 0.34 grams DCW/hr could be produced, around six times higher productivity than that with the original batch cultivation. Also, the target enzyme productivity in terms of the activity could reach 19.17 U/L/hr, 5.6 times higher than that with the batch fermentation (3.44 U/L/hr).
