α-mangostin induces G1 cell cycle arrest in HCT116 cells through p38MAPK-p16INK4a pathway

초록/요약
The alpha-mangostin (a-MG), one of active substance in the Garcinia mangostana, has shown to have anti-cancer effect in various cancer cell types. -MG treatment induces G1 phase arrest in cancer cell model by cell cycle dependent kinase inhibitors (CDKIs) induction and subsequent loss of CDKs activity. However, the precise molecular machineries underlying effect of -MG on cell cycle arrest remains unclear. In this study, we observed that -MG inhibits the cell proliferation of HCT116 cells in a dose dependent manner. Of interest, although loss of p53 rescues -MG effect on cell cycle arrest as consistent as previous report, p21Cip1 expression was only marginally delayed in the absence of p53 after -MG treatment. Instead, we demonstrated that activation of p38 mitogen activated protein kinase (MAPK) also contributes to induce p16Ink4a, which is responsible for G1 cell cycle arrest by -MG treatment. These findings indicate that -MG has cytostatic effect on the colon cancer cell by inducing G1 phase cell cycle arrest through p38 MAPK-p16Ink4a axis.
