Isolation and characterization of a novel agarase-producing bacterium, Pseudoalteromonas spp. from guts of spiny turban shells

Several agar-degrading bacteria were isolated from gut of spiny turban shells. These bacteria were examined by Lugol’s iodine and identified as extracellular agarase-producing bacteria. Among them, one species made clear depressions around its colonies. According to 16s rRNA identification, it was identified as one of Pseudoalteromonas species and named as Pseudoalteromonas sp. JYBCL1. Agar-degrading ability of Pseudoalteromonas sp. JYBCL1 was examined by measuring viscosity-change of the inoculated medium. The viscosity of the inoculated medium was decreased by more than 60% after 20 hour-cultivation while pure medium did not show any decline of viscosity. The agarase produced by the isolate had optimum activities at 35℃ and pH 7. It had extremely strong resistance to ionic stress, for its activity was maintained more than 90% in 1000mM of NaCl concentration. In addition, it could saccharify raw Gelidium amansii directly and the efficiency was about a half compared to the efficiency of agar-saccharification.
