Chemostat culture of Methylomonas sp. DH-1 to investigate methane assimilation characteristics

Methylomonas sp. DH-1 is a type I methanotroph that uses methane as a carbon source. This bacterium is one of the brightest and most promising strains near future in that it can use methane gas, one of the harmful green-house gases that cause global warming. In this study, Methylomonas sp. DH-1 was cultured in 5 L fermenter with chemostat culture, one of the continuous cultures. Chemostat culture is a device that constantly regulates productivity of microorganisms with maximum cell concentration under each condition. By comparing the maximum concentration and productivity of cell under various conditions, the difference in cell activity was studied. Here, the effect of agitation speed and dilution rate on cell productivity and concentration was confirmed in chemostat culture. In addition, the characteristics of methane assimilation of cell depending on the culture characteristics were investigated. In the chemostat culture, methanol bioconversion from methane were carried out using cells harvested under each condition. The difference in methanol production by cell activity was confirmed. These results show that culture characteristics is important to improve the productivity of compounds synthesized using microorganisms.
