Diagnosing Alzheimer's disease through detection of tau protein in human blood using ultrasensitive nanogap sensor : from CSF to Blood

In this study, ultra-sensitive nanogap sensor was developed for detection of low concentration of tau to diagnose Alzheimer’s disease (AD). Recently, biomarkers in human blood have been extensively researched for the diagnosis of AD which is minimally invasive and inexpensive. Since biomarkers in blood have much lower concentrations than in CSF (Cerebrospinal fluid), many efforts for developing highly sensitive sensor have been performed. Highly sensitive EIS (electrochemical impedance spectroscopy) sensor with nanogap was developed and investigated for detection of total, O-g(O-glycosylated) and P(Phosphorylated) tau. The measured impedance values were analyzed by Impedance change rate because of minimizing the error caused by device variation. And total tau, O-g tau and P tau measurement from various samples such as cell lysates, mouse brain lysates, mouse blood, Human blood and CSF. These results demonstrate that the nanogap sensor have enough sensitivity for detection of total tau, O-g tau and P tau to diagnose AD using human blood.
