Signal-Enhanced Enzyme-Linked Immunosorbent Assay using Enzyme Bomb for Detection of Human Thioredoxin-1 as a Breast Cancer Marker

초록/요약
Since the dietary life of people became westernized, there has been an upsurge of patients who suffer from breast cancer. However, no proper detection kits for it are available due to a lack of sensitivity of its currently known biomarkers and a low contrast between the true and false positive signals of probes. Therefore, we developed the enzyme bomb which is able to encapsulate HRP as much as a half billion of molecules and includes anti-Trx1 antibodies for simple and high signal detection via ELISA. As a result, enzyme bombs were formed as stable probes with a size of 100nm and encapsulation amount up to 5.5x108 molecules per single particle. They could detect Trx1 with higher optical signal than existent nanoscale probes conjugated with few HRP molecules. Also, Trx1 was detectable from few femtomolar to nanomolar range. Hence, this research demonstrates that enzyme bomb has potential to be the main tool of breast cancer detection with enhanced sensitivity and signal intensity.
