A Rapid Detection Using Quantum Dots-Encapsulated Magnetic Bead

This thesis proposes the development of an aptamer based assay employing the quantum dots (QDs)-encapsulated magnetic bead. Traditional assay usually involves multiple washing steps of significant time and labor cost. In order to improve these problems, I proposed a single-step assay process that can be integrated in microfluidic systems. Quenchers and specific aptamers were integrated on the QDs-encapsulated magnetic bead for single-step detection of protein. Real-time detection was confirmed by observing that thrombin binding reaction was completed within 10min after thrombin addition. The quantitative target protein was analyzed by the change of fluorescent intensity according to the concentration of protein. My new approach is expected to enable the rapid and specific protein detection with reduced costs and minimized materials consumption.
